Isolation of three new arylpyrrolidones from Justicia secunda VAHL

Abstrakt

Justicia secunda VAHL belongs to the family of Acanthaceae ETHNOMED: VERWENDUNG and is a barely examined species. The aim of the study was to investigate J. secunda in phytochemical aspects and find unknown compounds. A methanol extract of leaves was partitioned subsequently with light petroleum and dichloromethane. The dichloromethane fraction was subjected to column chromatography on Sephadex LH-20 and silica gel. For the isolation of pure compounds, semipreparative HPLC was conducted on RP-18e to yield a pure substance (1) and an equimolar mixture of diastereomers (2 and 3). MS and extensive NMR measurements led to the structure elucidation of these compounds.
The three novel arylpyrrolidones differing in the number of hydroxyl groups and their relative configuration are named secundarellone A (1), B (2), and C (3). They are the first pyrrolidones isolated from the genus Justicia and even from the family of Acanthaceae.
